Investigation of osteocyte cyto-architecture through bio-imaging approach o Histology: fixation, staining, plastic embedding, sectioning, polishing, mounting o 3D Confocal imaging of osteocyte and reconstruction using Volocity, Amira and software o TEM sample preparation and TEM imaging o Statistical analysis among various murine genotype and disease type using Graphpad, Origin, SAS software 2). Bone-muscle crosstalk: imaging and quantifying solute transport across periosteum o Refined surgery and dissection skill on mice o Confocal multi-track time-series x-z scanning to track the penetration of tracer inside the periosteum vs time o Image processing using ImageJ, Matlab, Adobe Photoshop software 3). Investigation of osteocyte pericellular matrix using a novel dual-colored fluorescence recovery after photobleaching (FRAP) approach o Matlab modeling to simulate the experiment: solute transport in the matrix o Fluid shear force and drag force quantification o Fluorescent dye conjugation with protein o Confocal imaging synchronized with mechanical loading:Confocal multi-channel multi-time imaging & Bose mechanical testing system o Image processing: Matlab programming 4). Mesenchymal stem cells isolation and cell culture 5). On-going project: ex-vivo intracellular calcium signaling--
