Research Associate 04/16/2015 - current
Singapore Eye Research Institute, Singapore, Singapore Singapore
Industry: Healthcare (medical research)
Working as a Research Associate in Singapore Eye Research Institute on various Research Projects
Project 1: Investigation of LGR5 as a potential retinal stem cell marker To characterize the expression of LGR5 in the adult mammalian retina with the intention of understanding the potential of LGR5 to aid in retinal stem cell isolation for therapeutic purposes. Responsibilities 1. To characterize expression of LGR5 in the adult mammalian retina • Identification of specific retinal cell type expressing LGR5. Analysis of anatomical distribution of LGR5 positive cells in the retina (identification of novel retinal stem cell niches). 2. To investigate the plasticity and regenerative capacity of LGR5 positive cells in the retina • Analyze proliferation, self-renewal, clonal expansion and response to injury in vitro and in vivo. • Assess ability to differentiate into retinal neuronal precursors under appropriate culture conditions. Project 2: Dental Pulp Stem Cells (DPSC) as potential alternative source of retinal stem cells Responsibilities 1. Isolation and establishment of human dental pulp stem cell cultures from extracted teeth 2. Differentiation of DPSC and Gene expression analysis of differentiated DPSC 3. Analysis of retinal neuronal functionality of differentiated DPSC in vitro Project 3: Differentiation of Primary Mouse Muller Glial towards Retinal Progenitor fate In Vitro Responsibilities 1. Establishment of primary muller cell cultures from mouse retinae and human retinae 2. Evaluation of expression of ASCL1 in primary mouse Muller cells qRT PCR, IHC and Western blotting.--

Cancer Science Institute, Singapore, Singapore Singapore
Industry: Healthcare
Worked as a Researcher on identifying the causes of DNA methylation aberrations in tumor cells.
1. To delineate the functional aspects of P53 in DNA methylation & consequences of P53 mutation in cancer. 2. To identify genes and pathways that are regulated by TP53-DNMT1 complex and rescue the epigenetic role of TP53 in TP53 altered cells. Responsibilities •Identifying hypometylated CpGs linked to p53-mutations that correlate with gene repression. •Characterize p53-DNMT1 target genes by identifying p53 & AP1 binding sites in p53 defficient and Isogenic cell lines using CHIP. •Preparation of Libraries for CHIPseq (High-throughput sequencing - MiSeq). •Characterization of methylated cytocines on the promoters using bisulfite sequencing. •Determining methylation status of AP1 binding sites using Illumina infinium 450k platform.--
